产品介绍
一. 制备 Dil染色液
a.制备 DMF、DMSO 或乙醇储备溶液：储备溶液应在二甲基甲酰胺 (DMF)、二甲基亚砜 (DMSO 或乙醇 DMSO 中配制，浓度为 1-5 mM。溶剂DMF 优于乙醇。储备溶液分装冷冻至少 -20℃。不能反复冻融，尽量一个月内用完。
b. 制备工作溶液：将储备溶液稀释到合适的缓冲液如无血清培养基、HBSS 或 PBS 配制成 1 至 5 μM 的工作液。工作液需要现配现用。
二.染色
1） 悬浮细胞
a. 细胞悬液1000g 4℃离心3-5分钟，弃上清，PBS洗2次，每次5分钟。细胞密度为1×106/mL。
b. 加入1 mL Dil工作液，室温孵育5-30分钟。
c. 4℃，400g离心3-4分钟，弃上清。用PBS洗涤2次，每次5分钟。
d. 用无血清细胞培养基或PBS重悬细胞，荧光显微镜或流式细胞术观察。
2）贴壁细胞
a．在无菌盖玻片上培养贴壁细胞。
b.从培养基上取下盖玻片并吸出多余的培养基。
c.加入100 μL工作液，轻摇使之完全覆盖细胞，室温孵育5-30分钟。
d.用培养基洗两次，每次5分钟。通过荧光显微镜或流式细胞仪观察。
此方法为荧光染色常用方案，仅供参考。
