K +通道阻塞;K + channel blocker;Tolbutamide is a sulfonylurea agonist for secretion of insulin by beta-cells. In rat pancreatic beta-cells cultured in the presence of Tolbutamide displayed increased numbers of beta-cells in a dose responsive manner with beta-cell numbers doubling at 100 μg/ml of Tolbutamide. The compound displayed inhibitory action towards beta-cells at concentrations greater than 100 μg/ml. In addition in the presence of low glucose concentrations Tolbutamide’s cytotrophic action was inhibited. On isolated dog pancreas islets Tolbutamide at 180 μM stimulated the release of somatostatin, insulin and glucagon in the absence of glucose. in all situations the loss of Ca2+ eliminated all effects of Tolbutamide. 体内研究：

Tolbutamide (125 mg/kg，在日常饮食中补充 27 周) 降低非肥胖糖尿病小鼠的糖尿病发病率。
Tolbutamide (100-400 mg/kg，腹腔注射，第 13 天给药 400 mg/kg; 第 10-13 天给药 100 mg/kg; 联合用药: 第 10-12 天 100 mg/kg，第 13 天 400 mg/kg) 降低孕鼠胎儿死亡。
