PROTAC BRD9 Degrader-7 is an orally active, selective BRD9 PROTAC degrader (DC50 = 1.02 nM). PROTAC BRD9 Degrader-7 mediates BRD9 degradation via the ubiquitin-proteasome system. PROTAC BRD9 Degrader-7 exhibits proliferation inhibitory activity in the MV4-11 cells. PROTAC BRD9 Degrader-7 can be used in the research of acute myeloid leukemia and other related malignancies. (Pink: BI 7271, Blue: 5-Aminothalidomide, Blue + Black: Thalidomide-5-piperazine, Black: N,N'-Dimethylpiperazine).

体外研究：

PROTAC BRD9 Degrader-7 (Compound C6) (1-100 nM) 在 MV4-11 细胞中表现出强效 BRD9 降解活性，10 nM 时降解率为 93%，100 nM 时降解率为 99%。
PROTAC BRD9 Degrader-7 (0.0045-10 nM，0-24 小时) 在 MV4-11 细胞中以剂量和时间依赖性方式降解 BRD9。
PROTAC BRD9 Degrader-7 (7 天) 对 MV4-11、MOLM-13 细胞表现出细胞毒性，IC50s 分别为 3.69、21.03 nM，对 HEL、K562、HL-60 细胞无细胞毒性。
PROTAC BRD9 Degrader-7 (0.1-100 nM) 不会诱导 MV4-11 细胞中 BRD4 和 BRD7 蛋白的降解。
