Retosiban (GSK221149A) is a potent, selective, and orally active oxytocin receptor antagonist (Ki (hOT) = 0.65 nM, Ki (rOT) = 4.1 nM) with no detectable agonist activity. Retosiban has nanomolar affinity for the oxytocin receptor with >1400-fold selectivity over the closely related vasopressin receptors. Retosiban inhibits spontaneous and induces uterine contractions. Retosiban can be studied in research for preterm labour.

体外研究：

Retosiban (Compound 22) 在三种临床前物种 (大鼠、犬、食蟹猴) 的微粒体中具有低至中等的固有清除率[1]。
Retosiban (1 nM-1 μM，静脉注射，单剂量) 在麻醉且非妊娠的 Sprague-Dawley 雌性大鼠中产生剂量依赖性催产素诱发的子宫收缩减少，ID50 = 0.27 mg/kg 和 IC50 = 180 nM[1]。
Retosiban (Compound 74) (0.3-3 mg/kg，静脉注射，单剂量) 以剂量依赖性方式显著降低妊娠晚期大鼠的自发性子宫收缩[3]。
Retosiban (Compound GSK221149A) (0.1-1 mg/kg，静脉注射，单剂量) 可剂量依赖性地减少麻醉大鼠中催产素诱发的子宫收缩[4]。
Retosiban (5 mg/kg，口服，每日一次，连续 1 天和 4 天) 可显著抑制未产雌性 Sprague-Dawley 大鼠中催产素诱发的子宫收缩，抑制率达 72%[4]。

体内研究：

Retosiban (Compound GSK221149A) (0.1-10 μM) 导致大鼠离体子宫肌层条带中催产素诱导的浓度反应曲线平行右移。
Retosiban (1 μM) 可减轻高张力下拉伸对组织子宫肌层收缩力的影响。
